With POP3 email, shouldn't the TO field auto-fill the rest of a contact, if
that email address is in Contacts in Outlook? If it doesn't, is it just a
case of the address needs to be entered manually once or twice before it's
cached?

The latter. The autocompletion cache remembers the last 2,000 addresses to
which you've sent mail. It does not reference your contacts in any way
whatsoever.
